Firstly, you will find a lot of debate as to whether you should set off with water in your tank or not. Some people will travel with an empty tank, fill up when they reach their campsite and empty it when they leave.
I know that the Zig water gauge had a calibration knob where you filled the tank and then adjusted the water gauge to read full. This was not very accurate and required setting up every time you filled up.
I wouldn't worry about it. Put some water into the tank and top up as necessary.

Speaking as someone who also has a Bailey I'd say it's worth doing if you're going to fill from an Aquaroll, simply because the system cuts out when it senses it's 100% full whether it is or not, you could find yourself with less water than you expected which may or may not be a problem depending on how you camp or how often you want to do refill runs.
If you're filling from a hose, just fill it till it overflows.
The calibration should be done with the tank empty.
