As mentioned send a invoice and that then starts a timer for you, Generally ebays rule is 7 days but you can open a NPB dispute much sooner, thats up to you at what point, personally I wait a max of 3 days without contact, then a further 4 days close the case, add the buyer to your banned bidder list (essential) and either offer a second chance or relist, You must use the tools ebay provide you with to ensure you get your fee's back, the item gets back for sale asap and the buyer gets a strike that will eventually stop them doing this to others, Of course they may just be away for the weekend and they'll find they've won and pay you
Biggest advice though is to send it SPECIAL DELIVERY no other service should be used..
