Rather begs the question, 'what is it about the A54 that jangles your nerves?'.
It's not a road I know, but looked at it on Google streeview and it seems just like any other road across that sort of terrain, and I've towed on plenty similar without much concern. If we're to offer alternatives (if such exists in terms of driving experience!), then rather need to know what you wish to avoid/minimise!
I've towed to a couple of sites in the region, one just off A523 south of Leek, and one off the A515 at Biggin, don't recall any issues. Only comment would be A515 through Ashbourne is a tad tight and wiggly in a couple of places, and you may need to do a hill start on the steep hill north out of town!
Maybe my nerves are less easily 'jangled' than yours, so I may not be seeing the things that concern you.
You ask about the A54 as an alternative to the A54! - I take that to be a typo, and maybe you meant A53!
Rather looks to me like you are not going to find much better routes, one is up to Macclesfield with 'town roads' then much the same country roads you seek to avoid on A537 across towards Buxton. The other route would be down the A523 past Leek to pick up A53. Neither for any obvious great gain as far as I can see.
