It's a good plan. If you want a stopover between Dublin and Kerry, try The Apple in Tipperary (also listed by someone else earlier) which is pretty much halfway between the two, and beside the wonderful Rock Of Cashel. In fact if I were pushed my recommended highlights of Ireland tour would be Dublin / the gardens of Wicklow, then the historical areas of Tipperary / Kilkenny and finally the sea and mountains of Kerry.
Quickest route to Killarney from Dublin won't take you near the apple, I'm afraid. It will take you through Adare which is very pretty and has a lovely Wee site.
There is a toll at Portlaoise heading south. Avoid the N7 in rush hour. It is a pita. You will be passing citywest, where I work.
