If you mean the Western side of France - St Jean de Luz, Biarritz - Atlantic coast then I'm not familiar with that area so will leave suggestions to someone else.
However, if you mean the Mediterranean side of France/Spain, then I think the best way, and others agree, is Calais, Abbeville, Rouen, Chartres, Orleans, Vierzon, Clermont Ferrand, Millau, Montpellier - and then in whatever direction along the Mediterranean!
It's the cheapest in terms of tolls, and the most spectacular in terms of scenery. There are lovely places to stop along the way.

You say southwest of France, so I take it to mean somewhere on the Atlantic coast southwest of Bordeaux.
Well, we are allergic to paying tolls on motorways, and have all the time in the world, so we go via Abbeville, Rouen, then either via Evreux, Dreux, Chartres, Tours, Poitiers, Angouleme, Bordeaux, all via the N10 which, south of Tours is an excellent road, often motorway standard. Or, we may go Abbeville, Rouen, Alencon, Le Mans, then Tours and as above.
If you are in a hurry and don't have the luxury of time, then of course the motorway is the quickest, but as we have never done it that way, perhaps someone else will advise of the best route.
