M6 route would leave you the wrong side of Derby for Ripley, you'll have to follow the A38 thru Derby. Easy towing tho - all dual carriageway, with a fairly big hill just north of Derby. (It's about an hour from Stoke to Derby, plus Derby to Ripley = another 20 mins).
From Stockport I used to go via Chesterfield to Ripley, granted I wasn't towing. The A6 from Stockport all the way through from Stockport is another option.
Think the best route from Bolton is probably the A628 (Woodhead?) and down the M1.
Enjoy. It's a lovely site, although buzzed a little by the police helicopter. Presuming you are staying at Golden Valley.
Definately M62 then M1, off at junction 28, A38 signposted Alfreton, then continue on past Alfreton and follow the signs for Ripley via the A610, first left off roundabout. 2nd left takes you into the town centre.
We live a couple of miles down the road lol

Cheer guys thanks for the help looks like the M62 is the quickest option, will have a good look at the route in the morning to workout how long it should take
M62/M1 may be quicker but it adds ten miles to your journey compared to going over Woodhead - I've done both routes when going to/from Norfolk and prefer Woodhead, but it's a twisty winding road full of HGVs, so preferable to go really early in the morning to avoid the majority of them.

Maybe they do, but there's not as many of them to hit you going round the bends!

Guys thanks for the advice we went over woodhead going 9am on Good Friday and came back M1/M62 on easter Monday, both took around 2 hrs. and the site was brilliant
Glad you enjoyed it. What did you think of the drive over Woodhead? Some nice scenery if you're the passenger, if you're the driver then you don't see much of it! lol
